Materials
To prepare 1L of 1M HEPES buffer, you need:
- 238.3 g HEPES
- NaOH
- deionized water
Procedure
- Dissolve HEPES in about 800 mL of deionized water.
- Adjust pH to 7.5 with NaOH.
- Add deionized water to 1L.

I think you can sterilize it. A short exposure should not be a problem. If you want to be sure, maybe you can filter in the dark (protected from the light).

I was wondering if I should filter sterilize this after dissolving it? I know it can turn into hydrogen peroxide with light exposure, and the sterilization process does expose it to light more than not sterilizing it?

Hi Elisabetta,
You can store this buffer at room temperature, or at 4C. If you store it in the fridge, you have to warm it up to room temperature before you start using it. Generally with buffers it is possible that salts precipitate if buffers are stored at low temperatures. Then you may need to heat it up to redissolve the salts.
My recommendation would be, if you plan to use it at room temperature anyway, to store it at room temperature.

Which storage temperature is good for this buffer? I store it in the fridge, but I think that if I have to some experiment with it, I need it at room temperature. I'm having some problems in doing the baseline on my software, because it seems that if I analysed the another 500 μl of the same stock buffer solution, it seems a completely different buffer, and it starts absorbing
