10x TBS buffer (10x Tris-buffered saline)

Materials

To prepare 1L of 10x solution, you need:

• 60.6 g Tris
• 87.6 g NaCl
• 1M HCl
• deionized water

Procedure

1. Dissolve Tris and NaCl in about 800 mL of deionized water.
2. Adjust pH to 7.6 with 1 M HCl.
3. Add deionized water to 1L.
4. Store at 4°C.

Dilute stock solution 10:1 to make a 1x working solution.

1x TBS buffer will contain 50 mM Tris-Cl, pH 7.6, 150 mM NaCl.

  That is correct, 8.76 g for 1x solution, or 87.6 g if you prepare a 10x stock, like described in this protocol.

  150 mM NaCl is 8.76 g not 87.6g.
